Material guide: ambroxan — ambergris that no longer needs the whale

CAS 6790-58-5, the true face of "ambergris" in modern perfume. A 2023 review calls it the most prominent olfactive component of ambergris: industry long made it by semisynthesis from clary sage's sclareol, and now a 100% renewable route runs from fermented beta-farnesene through an engineered enzyme cyclisation. Three generations of production, with the whale stepping offstage. 61 suppliers, high strength with a 1%-or-less evaluation advice, a substantivity record past 400 hours.
The ambergris legend is perfumery's most retold story: a sperm whale's intestinal product drifts at sea for years, cured by waves and sun, a windfall for whoever finds it. The legend is true. But the "ambergris" you smell in a modern perfume almost never involved a whale: the lead is this synthetic molecule.
What matters
- Ambroxan is the core molecule of the ambergris smell, the pillar of modern amber and the whole "clean skin" direction. High strength, evaluate at 1% or less, with a substantivity record past 400 hours.
- Its production history is a story of the whale stepping offstage: from beachcombed ambergris, to semisynthesis from clary sage's sclareol, to a 100% renewable fermentation route. Same molecule, three generations of origin.
- The 400 hours sit at the source scale's ceiling (and this record was measured on a 10% dilution), meaning "very long" rather than a precise value. Buying it is buying a base note that all but refuses to leave.

Plain reading: CAS is the material's ID number. Two series records in one table: a 48-month shelf life (a first — stable enough that freshness races don't apply) and the >400-hour substantivity (the vanillin article explained 400 as the scale's ceiling, and this one was measured at 10% dilution besides). It is a crystal at room temperature; make a dilution first.
The whale steps offstage
In 2023, Eichhorn and Schroeder's review in the Journal of Agricultural and Food Chemistry laid the molecule's history out on one line:
(-)-Ambrox, the most prominent olfactive component of ambergris, is one of the most widely used fragrance materials. Historically it was made from the diterpene sclareol, modified and cyclized into (-)-ambrox by classical chemistry steps. […] An alternative approach uses fermented β-farnesene as feedstock, converted to (E,E)-homofarnesol and then enzymatically cyclized with an engineered squalene hopene cyclase into (-)-ambrox — enhanced sustainability and carbon efficiency as 100% renewable Ambrofix.
Sclareol comes from clary sage, and in 2012 biotechnology took over that step too. Schalk and colleagues in the Journal of the American Chemical Society:
Ambrox is produced industrially by semisynthesis from sclareol, a diterpene-diol isolated from Clary sage. We identified and characterized the enzymes responsible for sclareol biosynthesis and reconstructed the pathway in genetically engineered Escherichia coli, achieving sclareol titers of ~1.5 g/L in high-cell-density fermentation — a basis for an alternative, sustainable and cost-efficient route.
On a timeline: beachcombed ambergris (animals and luck), clary sage fields (agriculture), fermentation tanks (engineering). Set beside gamma-decalactone's yeast peach and rose oxide's yeast stereochemistry, fermentation is taking over fragrance production one material at a time. For a buyer it means the same catalogue name can come from different generations of route; the origin line and "natural" designations on the specification deserve a look, purchasing common knowledge.

Smelling it
- Evaluate at 1% or less, per the database. Its character does not shout: faintly salty, faintly sweet, a dry papery-woody "skin" smell. Plenty of first-time smellers say "isn't this just… the smell of clean?" — which is exactly why it conquered modern perfumery. Experience speaking.
- The modern usage is often structural and heavy: whole perfumes are laid on top of it, with everything else worn as clothing. As a beginner, work the other way: start below 0.5% as a primer and feel how it stretches a formula.
- With ethylene brassylate it shares the quiet-giant role: unimpressive alone, everything collapses when you remove it.
- "Tea" and "old paper" in the descriptors are no accident; diluted, that quiet dryness genuinely leans both ways.

Regulatory
Listed on GRAS, JECFA, CoE, FEMA GRAS, IFRA. As always, registers carry no limit values. Animal-derived ambergris tincture additionally runs into each country's rules on cetacean products (mostly beach-cast material, regulations vary by jurisdiction); synthetic ambroxan carries none of that baggage. Buying it
- One small bottle lasts a very long time: high strength, >400 hours, 48-month shelf life — the series' definitive buy-once material.
- Read the origin line on the specification: the sclareol semisynthesis and the fermentation route share a CAS, and the premium on "natural" or sustainability claims is yours to accept or decline.
- Make a 1% dilution, then enter formulas below 0.5%. Its presence surfaces slowly.
